Molecular Structure of Philadelphia Chromosome Positive Leukaemias

چکیده

The Philadelphia chromosome was the first specifıc karyotype abnormality associated with a particular neoplastic disease in humans. Ph translocation generats hybrid gene consisting of 5' exon sequences bcr on 22 fused to 3' exons and polyadenylation/termination ABL proto-oncogene from 9. molecular consequence this is transcription chimaeric m RNA transcribed novel 210 kd protein (p 210) CML, an p 190 ALL which retain tyrosine-kinase activity derived sequences. Although breakpoints chromoseme 9 can occur over 200 KB range breakpoint are usually clusteret within 5 sequence designated cluster region. some positive acute lymphoblastic leukaemias, however, break has been shown lie further such that only ofbcr includled RNA.

Philadelphia Chromosome Positive Childhood

In a 3-yr period, the Philadelphia chromosome (Ph’) was found in 4 of 43 children with acute lymphoblastic leukemia (ALL) in whom chromosomes were studied at diagnosis. The clinical, morphological, cytochemical, and immunologic findings in the Ph’-positive (Ph’ + ) cases were consistent with typical childhood ALL, indicating that identification of cases requires chromosome studies. A review of ...

Molecular heterogeneity of adult Philadelphia chromosome-positive acute lymphoblastic leukemia.

The (9;22) translocation which produces the Philadelphia (Ph1) chromosome activates the abl oncogene from chromosome 9 by recombination with the bcr gene from chromosome 22. This fusion gene is transcribed into a new 8.5-kilobase chimeric mRNA which is translated into a novel Mr 210,000 fusion protein which has a protein tyrosine kinase activity that is greatly increased in comparison to the ac...

Philadelphia chromosome-positive leukemias: from basic mechanisms to molecular therapeutics.

The Philadelphia chromosome translocation (t(9;22)) results in the molecular juxtaposition of two genes, BCR and ABL, to form an aberrant BCR-ABL gene on chromosome 22. BCR-ABL is critical to the pathogenesis of chronic myelogenous leukemia and a subset of acute leukemias. The chimeric Bcr-Abl protein has constitutively elevated tyrosine phosphokinase activity. This abnormal enzymatic activatio...
